Vol. 206 No. 3 595-606 www.jem.org/cgi/doi/10.1084/jem.20081385 595 Monocytes, macrophages, and DCs form networks of phagocytic cells throughout most tissues, the development of which are dependent on the CSF-1 receptor (csf1r, also known as CD115, c-fms, and M-CSF receptor) ( 1, 2 ). These cells, which are sometimes referred to as the mononuclear phagocyte system, play major roles in development, scavenging, infl ammation, and antipathogen defenses ( 3, 4 ). They are highly heterogeneous in phenotype, tissue distribution, and function ( 3, 5, 6 ). Considerable attention is currently focused on the characterization of their progenitors and precursors, the signals driving their development in the BM, their migration to tissues, and their homeostasis in peripheral tissues. CSF-1R and its two known ligands, M-CSF and IL34 ( 7 ), are critical for the development of this lineage because M-CSF – defi cient mice (op/ op and csf1 / ) have a milder phenotype than the Csf1r-defi cient mice ( 8 ). Other cytokines, such as GM-CSF, FLT3, LT1 2 (LT ) ( 9 – 15 ), and chemokines ( 16, 17 ) have also been shown to control the development and homeostasis of the macrophage and DC networks. Cellular cloning and transplantation studies have shown that many macrophage subsets, most of the conventional DCs (cDCs) in the secondary lymphoid organs of mice, and at least a fraction of the DCs in the mouse thymus probably originate from myeloid progenitors ( 18 – 20 ). CX3CR1+ CD115+ CD135+ common macrophage/DC precursors and the role of CX3CR1 in their response to inflammation
CX(3)CR1 expression is associated with the commitment of CSF-1R(+) myeloid precursors to the macrophage/dendritic cell (DC) lineage. However, the relationship of the CSF-1R(+) CX(3)CR1(+) macrophage/DC precursor (MDP) with other DC precursors and the role of CX(3)CR1 in macrophage and DC development remain unclear. We show that MDPs give rise to conventional DCs (cDCs), plasmacytoid DCs (PDCs),...
متن کاملCombinatorial model of chemokine involvement in glomerular monocyte recruitment: role of CXC chemokine receptor 2 in infiltration during nephrotoxic nephritis.
A sequential model involving chemokines has been proposed for leukocyte extravasation into areas of inflammation; however, site-specific aspects remain to be elucidated. Hence, we studied the role of chemokines produced by mesangial (MC) or glomerular endothelial cells (GEC) and their receptors in glomerular recruitment of monocytes. Stimulation of MC with TNF-alpha up-regulated mRNA and protei...
متن کاملAssessment of levels and health risk of heavy metals (Pb, Cd, Cr, and Cu) in commercial hen’s eggs from the city of Hamedan
Increasing consumption of chicken products in Iran makes it very important to analyze their residual heavy metal contents; therefore, the present study has been conducted to determine Pb, Cd, Cr, and Cu in commercial hen eggs, marketed in the city of Hamedan in 2016. In this descriptive study, a sum of 27 samples of hen eggs has been collected from the Hamedan City's market. After preparation a...

متن کاملDetermination of heavy metal (Cr, Zn, Cd and Pb) concentrations in water, sediment and benthos of the Gorgan Bay (Golestan province, Iran)
The coastal area of the Caspian Sea and Gorgan Bay are important ecosystems receiving discharge from their tributaries. In this study, concentration of lead (Pb), cadmium (Cd), zinc (Zn) and chromium (Cr) was seasonally determined at 8 sampling points during 2009-2010. Water samples were collected from the sampling stations and transferred to laboratory in polyethylene containers, whereas, sedi...
